Inhibition of Topoisomerase II α Subunit De Novo Synthesis by Specific Antisense Oligonucleotides Suppresses Human Gliomo T98G Cell Growth

本実験では様々な抗ガン剤の標的酵素とされるトボイソメラーゼIIに対するアンチセンスオリゴヌクレオタイドを用いて、T98Gヒトグリオーマ培養細胞の増殖を抑制し、その際トボイソメラーゼII酵素活性は低下しトポイソメラーゼI酵素活性は増強される傾向にあることを確かめた。このことよりトポイソメラーゼIIはT98Gヒトグリオーマ細胞が順調に増殖するためには不可欠な酵素の一つで、これを抑制することで細胞増殖を抑制し得るか、トボイソメラーゼIがこれに相補的に働き耐性を獲得するという機序が想定された。脳腫瘍の化学療法において、相補的に働く酵素群による耐性獲得を制御できればより効果的な薬剤使用法が開発されると期待される。
