Expression and Regulation of Bone Morphogenetic Proteins in Human Neoplastic Epithelial Cell lines

Human salivary adenocarcinoma cells, HSG-S8 induced ectopic bone formation in their tumors when injected subcutaneously and intramuscularly into the nude mouse. Thereafter, it was demonstrated that bone morphogenetic protein(BMP)-2 mRNA was expressed in HSG-S8 and its protein was secreted into the medium. By reverse transcription-polymerase chain reaction(RT-PCR), BMP-5 and BMP-6 mRNA were also expressed in HSG-S8 cells. The in vivo bone-inductive potential of HSG-S8 cells was probably due to BMPs secreted by HSG-S8 cells of the nude mouse tumor. The expression of BMP-2 in HSG-S8 cells was stimulated in dose dependent manner by retinoic acid and 1α, 25 dihydroxyvitamin D_3 and inhibited by the treatment of dibutyryl cyclic AMP. Moreover, several subtypes containing BMP-2 were expressed in other neoplastic epithelial cell lines such as squamous cell carcinoma and adenocarcinoma originating from gastrocolic tissues. Nevertheless, HSG-S8 solely induced ectopic bone formation and endochondral ossification in nude mouse tumor, although the reason of this fact has not yet been revealed.
