6. Effects of Transforming Growth Factor-b1 on decorin mRNA expression and alkaline phosphatase activity by rat osteoblastic cell lines.

TGF-b is stored in the bone matrix in abundant amounts and modulates the synthesis of bone matrix proteins by osteoblasts. The present study investigated the effect of TGF-bl on the expression of decorin mRNA and alkaline phosphatase activity in two clonal rat osteoblastic cell lines with different stages of differentiatioun. The TGF-bl effect on decorin mRNA expression of C26 cells was inhibited in dose dependently and time cource. However, decorin mRNA was not detected in C20 cells under basal conditions in the presence or absence of TGF-bl. Treatmeunt of both C26 and C20 cells with 0.1-5.0 ng/ml of TGF-bl for 24 h decreased alkaline phosphatase activity in these cells dose dependently.
