Replication of Xestia c-nigrum (Lepidoptera : Noctuidae) Nuclear Polyhedrosis Virus in Continuous Cell Cultures

Replication of Xestia c-nigrum nuclear polyhedrosis virus (XcNPV) T-3,cloned by plaque purification, was studied in Spodoptera frugiperda and Spodoptera littoralis cells. Insect cell cultures were infected with XcNPV T-3 and extracellular infectivity was titrated by TCID_50 and PFU. The extracellular infectivity titer of XcNPV T-3 in S. frugiperda cell cultures was higher than that in S. littoralis cell cultures.Sensitivity of in vivo and in vitro titration methods of XcNPV T-3 was compared and assay in Spodoptera litura larvae was most sensitive. Results of TCID_50 and PFU assays in S. frugiperda cells were highly reproducible.
