Axonal swellings in the cerebellar white matter of groggy mutant rats were examined with synaptophysin (SYP) immunohistochemistry at light and electron microscopic levels. The axonal swellings displayed several gradations of SYP immunostaining density at light microscopic level. Immunoelectron microscopy revealed that the axonal swellings contained various amounts of SYP-immunopositive substances which were frequently attached with small vesicles measuring about 50 nm in diameter. These results suggest that an impairment of fast axonal transport is involved in the pathogenesis of these axonal swellings, and that the SYP proteins accumulated in the swellings are digested at that place together with other cytoplasmic organelles.
