Protoheme and heme c contents in photosynthetically grown Rhodopseudomonas sphaeroides cells varied to a great extent according to culture conditions. The specific content of protoheme (heme/protein) decreased during the logarithmic phase, increased thereafter, and remained constant at the stationary phase, whereas the content of heme c was constant throughout the whole culture period. A low value of the specific content of protoheme was found in the media composed of peptone or glutamate and essential vitamins, but a high value was found in those composed of ammonium, peptone or glutamate and yeast extracts, while the specific content of heme c was constant in all the media used. Further constant contents of protoheme and heme c were found in the media composed of different carbon sources. By addition of Fe^<3+>, Co^<2+> and Cu^<2+> to the medium, the ratio of protoheme to heme c became lower. Each factor seemed to have different effects on the heme contents.
