<原著>ラット7,12-dimethylbenz(a)anthracene(DMBA)誘発乳癌におけるchemo-endocrine therapyの核DNA量解析に関する研究

In order to achieve a better therapeutic result in human breast cancer, a chemo-endocrine therapy was undertaken for DMBA-induced rat mammary carcinomas. The relation between the results of chemo-endocrine therapy and the changes of nuclear DNA content and biochemical estrogen receptor (ER) values was analysed in the mechanism of amelioration. One-hundred and one rats with DMBA-induced mammary carcinomas in total were treated in the following way ; 24 by ovariectomy (Ovx), 19 with mitomycin C (MMC) and 18 with tamoxifen (TAM) as the single treatment group, and 21 by Ovx+MMC, 19 with TAM+MMC as the combined treatment group. Before the treatment the nuclear DNA content histograms of the DMBA induced carcinomas showed a diploid pattern at the peak channel of 2.0 to 2.4c ; aneuploid (2.4〜3.8c) cells and over 4c cells, however, were found in 41.5〜44.6 % of the cancer cells. The incidence of aneuploid cells and over 4c cells decreased significantly after the treatment by Ovx or with TAM. After the treatment with MMC, the decreases of aneuploid cells and over 4c cells were less than after the endocrine therapy, while the anti-tumor effect was the same with Ovx. The combined therapy, group of Ovx+MMC or TAM+MMC, showed more significant decreases of aneuploid cells and over 4c cells than the group treated by Ovx or with TAM alone. Biological ER value was positive in all the cases before the treatment. The ER value was found to be decreased after the endocrine therapy, by Ovx or with TAM, but no significant changes of the value were found in the cases treated by chemotherapy alone as the treatment with MMC. Most remarkable decrease of the ER value was observed in the combination of Ovx+MMC. Thus it may be concluded that according to the relationship between the antitumor effects and the changes of ER value and DNA content, the endocrine therapy by Ovx and the chemotherapy with MMC have the effect on their respective target cells different from each other, and that the combination of Ovx and MMC leads to synergistic actions on the DMBA carcinoma with the highest anti-tumor effect.
