Fractionation of Galactosidase Relating to the Disappearance of Bombyx Humoral Lectin Activity

The result of SDS-PAGE analysis showed that the whole fat body gave a band corresponding to galactosidase, while any fraction obtained from the fat body of the domesticated silkworm, Bombyx mori, by means of gel filtration gave no band corresponding to galactosidase. Moreover, I researched on the activity of galactosidase in the fractions obtained from the fat body by means of gel filtration. The result showed that both of α-galactosidase activity and β-galactosidase activity were recognized in the whole fat body, while any activity was not recognized in the fractions. The result of FPLC Mono Q ion exchange chromatography on the fat body showed that the chromatogram revealed a peak at the salt concentration of 0.12 M. However, the result of the research of galactosidase activity showed that both of α-galactosidase activity and β-galactosidase activity were recognized on the fraction at the salt concentration of 0 M by Mono Q ion exchange chromatography. Namely, it seemed that the different result was obtained between the fat body and the haemolymph of B. mori.
