It is well known that photoaging skin histologically shows numerous and diverse changes in extracellular matrix components. These quantitative and qualitative alterations of extracellular matrix components are suggested to contribute to wrinkle formation. Although matrix metalloproteinase family is known as the proteinase which play key roles in the process of extracellular matrix degradation, serine proteinases which have proteolytic activity toward extracellular matrix components were detected in the media of human dermal fibroblast cultures exposed to reactive oxygen species. The stimulation of serine proteinases activity was observed in hairless mice skin exposed to ultraviolet (UV) A irradiation repeatedly too. Also, decrease of elasticity and collagen content observed in hairless mice skin exposed to repeated UVA irradiation were suppressed by the application of serine proteinase inhibitor in the period of repeated UVA irradiation. Based on these results, serine proteinases induced by UV or reactive oxygen species were suggested to play important roles in the wrinkle formation observed in photoaging skin.
