Using hybrid offspring developed through crossbreeding severely spontaneously hypertensive rats (M-SHRSP) with spontaneously hypercholesterolemic rats (SHC), it was possible to establish a colony of Hyper T-C rats exhibiting both hypertension and hypercholesterolemia. The present paper relates the development and characteristics of this new strain from generations F_1 through F_<20>. At 20 weeks of age, male Hyper T-C had blood pressures around 230 mmHg and plasma total cholesterol levels of approximately 400 mg/dl, for females at 25 weeks of age, the figures were about 230 mmHg and 500 mg/dl, respectively. Also, at this age, the plasma total cholesterol/high density lipoprotein (HDL) cholesterol ratio was relative low while that of the low density lipoprotein (LDL) cholesterol was noticeable high. Just as with SHC, Hyper T-C also manifested protein excretion in the urine, hypoalbuminemia and, under both light and electron microscopes, showed abnormal permeable lesions in the renal glomeruli capillary walls, findings similar to human nephrotic syndrome symptoms. It is thought that such increases in plasma total cholesterol levels are caused by these renal disorders. In addition to the above, Hyper T-C rats also showed evidence of typical hypertensive changes in a number of organs. For example, hypertensive changes in the afferent arterioles and resulting ischemic changes in the capillary basement membrane of the glomeruli were found in the kidneys. In particular, similar to human arteriosclerosis, there were arteriosclerotic changes found in heart and kidney muscular arteries with an outer diameter of 150 to 800 μm. The above findings show that these Hyper T-C rats have an excellent potential as an animal model for arteriosclerosis.
