光アフィニティーラベル化後修飾法によるレクチンの機能化

我々の開発した光アフィニティーラベル化後修飾法 (P-PALM) は、タンパク質の活性部位近傍に部位特異的に人工分子を導入できる優れた手法である。糖結合性タンパク質 (レクチン) に対してこの手法を適用することで、様々な機能化を行った。例えばP-PALMにより蛍光色素を導入することで、糖との結合を蛍光変化により読み出す蛍光性の糖鎖センサーを構築した。さらに、このセンサーに別の蛍光団を選択的に導入することで、測定精度を向上させ細胞内での糖鎖検出に成功した。またセンサー化のみならず、副結合部を導入することでレクチンの糖選択性の改変も行った。このようにP-PALMを用いることでレクチンを分子レベルで機能化することができ、新たな糖鎖解析のツールとして糖鎖研究にさらなる発展をもたらすことが期待される。
