All of the observations were made with preparations of Perilla leaf slices treated as follows:
1) fixation ; by means of gultaric aldehyde and osmic acid
2) dehydration ; by means of ethyl alcohol and propylene oxide
3) imbedding ; in epon resin
4) electronic staining ; by means of acetic-uranium and caustic-lead
In mesophyll cells, the authors observed the microscopic structures of nucleus, nucleolus, nuclear membrane, cell wall, cell membrane, cytoplasm, mitochondria, endoplasmic reticulum and chloroplast.
Within the cytoplasm of secretory cell, endoplasmic reticulum, mitochondria and osmium-stainable bodies were numerously found, but chloroplasts were not contained.
There was a close resemblance between the internal structure of secretory cell of oil glands and that of stalk cell. So it may be said that these two kinds of cells perform a similar function.
Small pits were observed in the walls of three kinds of cells, i. e. the basic cell, stalk cell and secretory cell. Through these pits, the cytoplasm of one cell connects with that of adjacent cells.
It seems that within the secretory cell the essential oil is synthesized from some kinds of materials which are produced in chloroplasts of mesophyll cells.
