Carbonic anhydrase inhibitor on the otolithic organs of developing chick embryos.

The purposes of this investigation are to observe the change of otolithic organ in the developing chick embryos after injection of carbonic anhydrase inhibitor, acetazolamide; and to compare the dose differences. Acetazolamide, each doses of 1.5mg, 3mg or 6mg/0.06ml/egg, was injected into the yolk sac of the embryo of 5 days of incubation. The specimen of embryos were collected on the 11, 13 and 18th day of incubation and on the 3rd day posthatching, and those were comparatively observed concerning abnormality in development of structure and posture, including the investigation using with SEM. There were the case with anopthalmos on the left side, or with exteriolized viscera, or with syndactyly of both 2nd and 3rd toes and opisthotonic spasm in the injected cases. A marked disturbance of otoconial formation; especially in reduced number and in some conglomeration and a widely exposed mesh-work structure of otolithic membrane were observed in many cases in the drug groups. There were several abnormalities, such as the absence of the otoconia in some cases, only single giant otoconium and/or markedly irregular size and shape of otoconia were seen. Conclusively, carbonic anhydrase inhibitor (acetazolamide) injected into the yolk sac of the developing chick embryos would act as the inhibition of both morphogenesis of the otoconia and formation of the otolithic membrane.
