Location of Potassium Ion Pump in the Stria Vascularis of Guinea Pig

The distribution of both DC potential and K-ion activity in the cellular layers of stria vascularis were investigated simultaneously by inserting a double-barrelled K-ion sensitive microelectrode into the scala media through the stria vascularis of guinea pig cochlea. The locus of the potassium ion activity observed with the advance of an electrode toward the endolymphatic space was irregular, while the positive DC potential seemed to be he intracellular potential of the strial cells increased always steadily. Considering from the DC potential differences and the changes of potassium ion activity which were recorded at each strial cell membrane, two points were remarkable to show very large electrochemical potential gradient for potassium ion. One is the marginal cell membrane of vascular side and the other is that of endolymphatic side. This result seems to suggest that two strong potassium ion pumps must lie in the marginal cell membrane of these both side respectively. In other words, it might be expected that the pump located in the vascular side draws in potassium ion to the marginal cell inside from the strial vessels and the another one located in the endolymphatic side draws out potassium ion into the endolymph from the cell inside.
