Ototoxicity of Polyamino-compounds (Report II):Their Effects on Phospholipids Metabolism of Brain Slice

We have previously shown that some polyamines had ototoxicity when perilymphatically perfused. Aminoglycosidic antibiotics are known to interfere with the metabolism of polyphosphoinositides of the inner ear. In order to examine the possible involvement of ototoxic polyamines in polyphosphoinositides metabolism, guinea pig brain slices were incubated with or without polyamines and their effects on <SUP>32</SUP>P incorporation into phospholipids were estimated. Polyamines employed were kanamycin, polymyxin B, poly-L-lysine, putrescine, and spermine.<BR>Rapid labelling of phosphatidylinositol diphosphate in the control study and its marked supression by the polyamines (10<SUP>-3</SUP>M and 10<SUP>-2</SUP>M) were observed. The degree of this supression, however, was not necessarily coorelated with their reported degree of the supression of cochlear microphonics by perilymphatic perfusion of these drugs, requiring further study.
