In Vitro Developmental Capacity of Rat Embryos Recovered at Various Times after Mating.

Wistar-Imamichi rats of 10 to 14 weeks old were housed under a controlled illumination (lights on 0700-2100 hr; hr). Embryos of various stages were recovered between 36 and 84 hr after mating (0 hr = 0500 hr on the day of estrus) at 3 or 6 hr intervals, and cultured in modified Krebs-Ringer solution for 12 to 36 hr. Two-cell embryos had no evidence of development when they were recovered at 36 to 48 hr after mating, but 45.1 and 67.6 % of them developed into the 3-and 4-cell stages at 54 and 57 hr-recovery, respectively. Of the 4-cell embryos recovered at 66 and 69 hr, 36.5 and 46.5 % developed into 5-to 8-cell stages after 12 hr-culture, respectively. When the 8-cell embryos were recovered at 75 to 81 hr after mating and cultured for 12 hr, the incidence of developing embryos raised from 18.1 to 65.4 % with the lapse of recovery time after mating, and about the half of them progressed to blastocyst after 24-and 36 hr-culture. At 84 hr-recovery, almost all of compacted embryos developed into further stages and 63.8 % of them reached to blastocyst after 24 hr-culture, but no hatched blastocyst was observed. These results suggest that the incidence of development of rat embryos raised with the lapse of recovery time and this shows that their embryos may be viable during the short period in modified Krebs-Ringer solution.
