The developmental potential of bisected frozen-thawed mouse embryos using three cryoprotective agents (glycerol, DMSO and ethylene glycol) was studied. Compacted morulae were bisected by the method of NAGASHIMA <I>et al</I>. (1984) and cultured in BMOC-3 for 10 hours. The embryos were then frozen and thawed with the three different kinds of cryoprotective agents. Normal zona free compacted morulae were frozen as the control.<BR>Morphological examination was done on the thawed embryos after 48 hours of in vitro culture. The percentages of bisected embryos that developed into normal blastocysts were 27.3% (glycerol), 34.2% (DMSO) and 39.4% (ethylene glycol). While, in the zona free embryos, the percentages were 31.5%, 37.5% and 41.3%, respectively.<BR>Thawed embryos that developed into normal blastocysts after 24 h of in vitro culture were transferred into recipients. The percentages of bisected embryos that developed into fetuses 7 days after transfer were 14.3% (glycerol), 19.0% (DMSO) and 28.6% (ethylene glycol). While, in the zona free embryos, the rates were 25.0%, 28.6% and 42.9%, respectively. However, there was no significant difference among the three cryoprotective agents in the survival rates of the bisected and the zona free embryos.
