The activities of cytochrome oxidase, NADH<SUB>2</SUB> dehydrogenase, succinate dehydrogenase, lactate dehydrogenase and Δ<SUP>5</SUP>-3β-hydroxysteroid dehydrogenase were histochemically examined in 8-cell mouse embryos frozen and thawed by the method of WHITTINGHAM <I>et al</I>. (1972) and in those cultured sub-sequently into morulae and blastocysts. The results obtained were as follows.<BR>1. Frozen-thawed 8-cell embryos were divided into three groups: embryos wholly composed of healthy blastomeres (Group I), those composed of both healthy and degenerated blastomeres (Group II), and those composed of only degenerated blastomeres (Group III). Healthy blastomeres from Groups I and II showed no enzyme activity except for a weak one of cytochrome oxidase. Degenerated blastomeres from Groups II and III showed abnormally strong activities of all the enzymes examined.<BR>2. Recovery in the enzyme activities was seen as follows: that of NADH<SUB>2</SUB> dehydrogenase in the morulae cultured for 24 hours and developed from wholly healthy frozen-thawed 8-cell embryos (Group I), and that of lactate and Δ<SUP>5</SUP>-3β-hydroxysteroid dehydrogenases in the morulae and blastocysts cultur-ed for 48 hours, and that of succinate dehydrogenase in the blastocysts cultured for 52 hours.<BR>These results suggest that the length of time needed for the recovery of metabolic ability in frozen-thawed embryos causes much a delay in the development of embryos.
