Role of sodium, potassium and protein in the occurrence of crystal configuration of bovine cervical mucus dry smear (CDS) was investigated.<BR>The configurations were grouped into fern-leaf-shaped, needle-leaf-shaped and non-figure-shaped, which appeared in the standing estrus, the pre-and post-estrus, and the diestrus, respectively. Observation by scanning electron microscope (SEM) showed that the crystal configurations consisted of small particles. Analysis with X ray analyser showed that sodium and chloride existed more distinctly than did potassium in the crystal configuration of bovine cervical mucus. The concentra-tion of sodium in mucus was higher in the standing estrus than in the diestrus, while the change in protein content was the reverse. Crystalization was induced with NaCl and KC1 in the presence of bovine serum albumin (BSA) solution, which was adjusted to the protein concentration in the mucus in the standing estrus and two days after estrus. The induced crystals, particularly made with NaC1 and BSA solution, showed a similar figure to the bovine CDS under the light microscope and SEM. The crystal did not occur after dialyzation of the mucus, but addition of NaC1 to the dialized one resulted in reappearance of the fern-leaf-shaped crystals.
