1) Effects of <I>in vitro</I> aging on the DNA content of spermatozoa were studied with ejaculates from three Holstein bulls. The measurements of DNA were carried out cytophotometrically, using sperm in the semen stored at 5°C as control.<BR>2) The content decreased in the dead sperm after 5 hr. incubation at 50°C, but no change occurred in the cold shock (with dry ice).<BR>3) 35 hr. incubation at 37°C, both aerobic and anaerobic resulted in the different effect on the sperm DNA according to the initial amounts or the source of spermatozoa. There was no appreciable change in the bull [A] showing the highest initial content of sperm DNA, but it decreased in the bull [C] with the lowest content. In the bull [B] with the intermediate content, DNA decreased by the aerobic incubation.
