Varietal Differences in in vitro Plant Regeneration Ability in Bangladesh Chili Pepper (Capsicum annuum L.)

From the practical viewpoint of chili breeding using tissue culture technology in Bangladesh, <I>in vitro</I> plant regeneration ability and optimal culture conditions were investigated using six accessions derived from three indigenous chili pepper (<I>Capsicum annuum</I> L.) cultivars, Deshi Morich, Bograr Morich and Dhania Morich, which are most popular in Bangladesh. As explant sources, hypocotyl, cotyledon, cotyledonary node, first leaf and shoot tip were cultured on four shoot formation media, MURASHIGE and SKOOG (MS) using growth regulators. Significant differences in shoot formation were observed among the cultivars, explant sources and shoot formation media. Accessions of Dhania Morich showed a higher shoot formation ability than others. Meristematic explants (shoot tip, cotyledonary node) displayed a higher shoot formation ability than the non-meristematic ones. The MS medium with BA 5.0 mgl<SUP>-1</SUP> was optimum for shoot formation. Root formation was observed on the three media. The MS medium containing NAA 0.1 mgl<SUP>-1</SUP> and IBA 0.05 mgl<SUP>-1</SUP> led to the production of potential roots with vigorous shoot growth and easy acclimatization. After hardening, the plantlets were transplanted to the field and grew into mature plants. The best cultivar Dhania Morich and <I>in vitro</I> technique developed here will be used for the chili breeding programs using tissue culture in Bangladesh.
