We have cloned four genomic sequences encoding trehalase by screening the genomic library using trehalase cDNA as a probe. One region (about 5.5kb), which was not covered by clones, was amplified from genomic DNA by LA-PCR. The gene sequence spanned near 19kb in length and was interrupted by one rarely long intron (13965 bp) inserted between 5′ non-coding region and coding region. The highly reiterated sequences were found at several regions in the intron. The protein coding region was not interrupted by any introns. Two putative transcription initiation sites were determined by primer extension analysis.The amino acid sequence of the Bombyx trehalase was compared with those of other organisms including E. coli, yeast and mammal. The phylogenetic relationship and evolution of the trehalases showed that these trehalases would be derived from a common origin.
