To clarify the anti-bacterial action as a defense mechanism in the digestive juice of the silkworm, the properties of anti-Streptococcus protein (ASP) and the effect of dietary conditions on the anti-bacterial activity of ASP were investigated. ASP was found in the precipitates by ammonium sulfate at the concentrations of 0-30% and 30-70%, but not 70-100%. A single peak of ASP in both precipitates was obtained by Sephadex G-75 column chromatography, whereas two active peaks were obtained by DEAF-cellulose chromatography. ASP purified by Sephadex G-75 chromatography was highest in larvae reared on mulberry leaves in spring; in the 175-time dilution the sample showed the activity to inhibit bacterial growth completely. In larvae reared on the leaves in later autumn, the 59-time dilution was effective. However, in larvae, which were reared on artificial diet by the end of the 4th instar and no leaves thereafter, only 18-time dilution was effective. When reared on the leaves stored for a long period, no dilution was effective. Thus, the presence of two forms is at least suggested and their production is seemed to be controlled by food condition.
