Nuclear DNA content of small cell carcinoma of the lung and its relationship to the cytomorphologic features.

The relationship between the nuclear DNA patterns of tumor cells obtained by brushing the bronchoscopy and cytomorphologic features of 18 patients with small cell carcinoma of the lung, was investigated. The nuclear DNA content was measured at 550 nm using a Microspectrophotometer. The DNA histogram pattern was classified into "type A" containing a higher propotion of G<SUB>0</SUB>G<SUB>1</SUB>-phase cells and "type B " containing a higher propotion of S, G<SUB>2</SUB> M-phase cells.<BR>The diameter of the nuclei was distributed to 10.1±2.0μfrom 10.1±1.9μ and did not differ between the two types.<BR>The nuclear chromatin pattern of the tumor cells with type B histogram pattern showed finely granular chromatin evenly distributed throughout the nuclei.<BR>However, the nuclear chromatin pattern of the tumor cells with type A showed finely granular chromatin and coarsely granular chromatin distributed sparsely throughout the nuclei.<BR>The small cell carcinoma with type B showed a good response tumor to chemotherapy whereas the case with type A showed a poor response tumor.<BR>There was a strong correlation between the nuclear DNA histogram pattern and the response to chemotherapy; however, the nuclear chromatin pattern was not statistically different from the response to chemotherapy.<BR>These results indicated that the nuclear DNA histogram pattern may be an indicator that can be used to speculate the clinical behavior of patients with small cell carcinoma of the lung.
