On the Nitrogen Compound Distribution and Amino Acid Composition of Nonprotein Fraction in the Feces from Germfree and Conventionalized Rats

To assess the role of the gastrointestinal microflora in the digestion, the nitrogen compound distribution in feces and particularly the amino acid composition in nonprotein fraction in them were analyzed on germfree and conventionalized rats., <BR>The germfree and conventionalized rats in experiments (weighing approximately 230g) were fed on autoclaved casein 18% or protein free diet.<BR>In the feces of germfree rats, more peptide-N, amino-N, amide-N and urea-N were excreted than in those of conventionalized rats, on the contrary true protein-N and ammonia-N were excreted more than in conventionalized rats. The differences of the amount of excreted ammonia-N between germfree and conventionalized rats on the protein free diet were not significant.<BR>On the casein diet, the main nitrogen compound in the feces of germfree rats were peptide-N and amino-N, but in those conventionalized rats were true protein-N and peptide-N. On the protein free diet, the main nitrogen compound in the feces of germfree rats were peptide-N, amino-N and true protein-N, whereas in those of conventionalized rats were true protein-N.<BR>The total and free amino acid composition (g/16g N) of nonprotein fraction in the feces were different between germfree and conventionalized rats.<BR>All the 15 amino acid determined (except His and Cys on the casein diet) in nonprotein fraction of the feces were at heigher level in germfree than in conventionalized rats (g/16 g N). On the diet experimented, the heigher proportion of Glu, Ser, Asp, Lys and Thr were observed in nonprotein fraction of the feces in germfree and conventionalized rats. The amino acid pattern (g/16g N) of nonprotein fraction of germfree rats fed on the casein diet was simillar to that of the protein free diet, whereas in that of conventionalized rats were different between the casein and the protein free diet.<BR>The results obtained suggested that the gastrointestinal microflora acts not only on dietary proteins, but also on endogenous proteins.
