The content of sterol in the unsaponifiable matter of lipids extracted from spices (34 kinds) was determined and was presented as the quantity equivalent to that of β-sitosterol (β-S). The sterol portion separated from the unsaponifiable matter by thin layer chromatography was analyzed by gas-liquid (GL) chromatography (<I>cf</I>. also our previous reports : Part 1, 2, 3 and 4). The GL chromatographic peak of the same relative retention time (R. R. T.) as that of cholesterol (Ch) was examined for identification by mass spectrometry (MS).<BR>1) The sterol content of spices was 0.08-2.56 mg in 1g of dried material. As for sterol composition, the content of β-S was the highest in all of the samples, though in celery, cumin and fennel the content of stigmasterol (St) was higher than that of β-S. All of the samples contained St and campesterol (C) with β-S, except that C was not found in clove and St was not detected in horse radish, laurel and mustard. Brassicasterol (Br) was contained in fenugreek, mustard and onion. Δ<SUP>7</SUP>-stigmastenolwas found in celery, coriander, dill and tarragon. Ch was detected in fenugreek, garlic, onion, paprika and red pepper. In cumin a GL chromatographic peak not yet identified, whose R. R. T. was 1.14 (relative to 1.00 of β-S) by chromatographic columns OV-17 as well as SE-30, appeared near to that of Δ<SUP>7</SUP>-stigmastenol.<BR>2) The peak, which appeared just after that of β-S and was always found in melons (Cucurbitaceae) as reported previously (Part 3), was ascertained to correspond to that of Δ<SUP>7</SUP>-stigmastenol.<BR>3) The peak previously reported to correspond to that of β-S in pumpkin, leaf-beet, white gourd shavings, cucumber, oriental pickling melon, wax gourd, bottle gourd, <I>Cucumis melo</I> var. makuwa, musk melon and Prince melon (<I>cf</I>. Part 3 & 4) was confirmed to be the peak for α-spinasterol (Sp). It was found that Δ<SUP>7</SUP>-stigmastenol was also contained in many of the samples containing Sp, includingspinach. Sp was the main sterol in Cucurbitaceae so far examined by us, except in water melon, and Δ<SUP>7</SUP>-stigmastenol was also contained. The main sterol of pumpkin seeds was revealed to be Sp, Δ<SUP>7</SUP>-stigmastenol and Δ<SUP>7, 24</SUP>-stigmastadienol. It would be full of interest in view of the sterol biosynthesis that Δ<SUP>7</SUP>-sterols were found in Cucurbitaceae.<BR>4) It was ascertaind by GLC-MS that all of the peaks of the same R. R. T. as that of Ch so farreported and in the present work (fenugreek, garlic, onion, paprika and red pepper) were coincident with the peak for Ch.<BR>So it is sure that Ch is widely distributed in plants, some of which, especially egg plant, shallot, onion and others, contain it in a fairly large amount.
