The hypocholesterolemic mechanisms of konjac mannan (KM), pectin (Pec) and carboxymethyl cellulose (CMC) in cholesterol-fed rats were studied. The hypocholesterolemic effect of KM was confirmed. Distribution of exogenous cholesterol was examined at 2, 3, 4 and 6hr after feeding 4g 0f the diet containing 1% of 4-<SUP>14</SUP> C-cholesturol and 0.25% of sodium cholate with or without polysaccharide supplements to the rats trained to consume the daily requirement of diet within 2hr.<BR>Exogenous cholesterol levels retained in the small intestinal wall, blood plasma and liver tissue were significantly lower in rats fed KM, Pec or CMC than in those fed the control diet. Absorption of cholesterol in preformed mecellar phase was not affected with KM when examined using Cori's method (<I>in vivo</I> sac method).<BR>From these results, it was suggested that the plasma and liver cholesterol depressing activity of KM would be due partly to a reduced absorption of ingested cholesterol through the inhibition of cholesterol solubilization. Several possibilities were also discussed.
