Determination of Alpha-Keto Acids in Etiolated Soybean Seedlings by Thin-Layer Chromatography

Changes in content of α-keto acids in black soybean seeds during their germination in the dark were studied. 2, 4-Dinitrophenylhydrazones of α-keto acids were separated by thin-layer chromatography and then determined colorimetrically.<BR>Glyoxylate, pyruvate, oxalacetate, α-ketoglutarate and α-ketoisocaproate were found in every parts of the etiolated seedlings, except in the hypocotyls of the 10 days old.<BR>Glyoxylate occurred in a relatively high concentration in cotyledons, and the content of this aldehyde acid in the embryos increased considerably.<BR>α-Ketoglutarate decreased gradually in the cotyledons, and this keto acid and oxalacetate could not be recognized in the hypocotyls.<BR>The seedlings germinating in the light, as compared with the etiolated seedlings, retained more pyruvate and α-ketoglutarate in cotyledons. These acids were much lesser in the growing tissues.
