1. Murexide used in combination with the double-beam spectrophotometer made it possible to directly record the quick changes of Ca ion concentraction in a suspension of relaxing vesicles, viz., isolated sarcoplasmic reticulum. 2. In the presence of Mg ions and ATP, Ca ions were instantaneously bound to the relaxing vesicles. 3. The effects of Ca binding inhibitor, such as anions, lower aliphatic alcohols and detergents, were studied. 4. The mechanism of Ca binding of the relaxing vesicles was discussed.
