To determine the response of insulin and glucagon to administration of amino acid, experimental studies were carried out in conscious dogs with previously implanted indwelling catheter in the pancreaticoduodenal vein, the inferior vena cava and the mesenteric vein<BR>Intravenous administration of one g per kg of a mixture of ten-amino acids to 16 dogs over a 60-minute period raised amino nitrogen to the mean level of 13.5mg%, mean pancreaticoduodenal venous insulin rose from 84 to 459μU per ml and glucagon from 1.1 to 2.7mμg per ml. There was no fall in blood glucose level during hyperinsulinemia induced by the amino acid administration.<BR>Intraduodenal administration of one g per kg of the amino acid mixture over a 45-minute period was followed by a rise in insulin and glucagon concentration in the pancreaticoduodenal vein Intraduodenal ingestion of amino acid did not cause measurable release of intestinal glucagon-like immunoreactivity into the mesenteric vein plasma.<BR>To gain insight as to the functional role of aminogenic hyperglucagonemia, the effects of hyperglycemia were studied by giving the amino acid mixture during a constant glucose infusion at the rate of 200mg per minute. Glucagon secretion was suppressed by hyperglycemia prior to and during the amino acid infusion, and plasma glucose concentration fell to 35mg%, suggesting that absence of aminogenic hyperglucagonemia permitted more intense cataglycemia.<BR>When aminogenic hyperglucagonemia was effectively prevented by the use of dogs in which the glucagon-containing pancreas had been resected, their cataglycemia exceeded that of the controls in which glucagon secretion was not inhibited.<BR>It is concluded that the administration of amino acid stimulates the secretion of both insulin and glucagon, and that the hyperglucagonemia induced by the amino acid administration might play some role in glucose homeostasis.
