Analysis of Skeletal Muscle Phospholipids in Mice with Muscular Dystrophy

Major phosphogylcerides of skeletal and heart muscles from mice (Strain 129/ReJ) with or without hereditary muscle dystrophy were separated by thin-layer chromatography, and composition of their fatty acids was determined by gas-liquid chromatography.<BR>1) Relative contents of phospholipid classes in the skeletal and heart muscles, liver, brain, lung, kidney and spleen were not significantly different between the normal and dystrophic mice, although, in some cases, the contents of neutral lipids (mainly triglyceride) were shown to increase apparently with muscle dystrophy.<BR>2) In normal mice, phosphoglycerides, especially, phosphatidylcholine (PC) and phosphatidylethanolamine (PE) from the skeletal and heart muscles were characterized by a very high content of 20-22 carbon poly-unsaturated fatty acids, in comparison with those from other tissues. The most abundant poly-unsaturated fatty acid was identified to be docosahexaenoic acid (ω3), and accounted for 30% and 40% of the total fatty acids in PC and PE, respectively.<BR>3) In dystrophic mice, it was found that a striking decrease of docosahexaenoic acid (ω3) in phosphoglycerides from the skeletal and heart muscles was concomitant with increases of saturated, monoenoic and dienoic acids.<BR>4) PC and PE from the skeletal muscles and livers were separated into 4 major subfractions; monoenes, dienes, tetraenes and hexaenes on AgNO3-treated silica gel thin-layer plates. In the normal mice, hexaenoic PC and PE predominated in the skeletal muscles (50% and 60%), and, in the dystrophic mice on the other hand, the relative proportion of hexaenes decreased greatly.<BR>These data suggest that the membrane systems of muscle tissues in mice are of a biologically unique characteristic and differ from those of liver and other tissues, although the physiological function of poly-unsaturated fatty acids is not fully known at present. In the dystrophic mice, it is suggested that there are significant alterations of the membranes of muscle tissues.
