Several aspects of purine biosynthesis and its regulation in animal tissues are briefly reviewed. One of the principal problems is whether each animal tissue is able to synthesize purine bases that are needed or some tissues are dependent, at least partially, on supply of purines preformed in other tissues such as liver. Although it seems that most, if not all, of mammalian tissues are provided with the capacity to synthesize purines <I>de novo</I>, this does not necessarily mean that the purine metabolism in a tissue is entirely self-dependent. Available evidence indicates that active transfers of purines between tissues actually take place. Although"salvage" enzymes (AMP pyrophosphorylase and IMP pyrophosphorylase) are likely to participate in this transfer mechanism, no experimental evidence is available as yet. Overproduction of purine bases in congenital defect of IMP pyrophosphorylase (Lesch-Nyhan syndrome) poses important problems to biochemists interested in this field. One of these is how the defect is related to synthesis of excess amounts of purines de novo. With regard to this point, the results of studies by Rosenbloom <I>et al</I>. were cited.
