We found recently that dibutyrylcytidine 3',5'-cyclic monophosphate(Bt<SUP>2</SUP> cCMP)stimulates neurite outgrowth in PC12 cells. In this study, we compared the stimulating effect of Bt<SUP>2</SUP>cCMP with those of nerve growth factor (NGF) and basic fibroblast growth factor (bFGF). NGF gradually increased the population of cells with neurites during a 5-day culture period, while the stimulation by Bt<SUP>2</SUP> cCMP or bFGF was highest 48 hr after plating. Staurosporine (SS), an inhibitor of protein kinases, inhibited the neurite outgrowth induced by NGF, but not that by Bt<SUP>2</SUP> cCMP. On the other hand, the stimulation of bFGF was enhanced by SS. The neurite outgrowth induction by combined additions of Bt<SUP>2</SUP> cCMP and each peptide was additive. These results suggest that Bt<SUP>2</SUP> cCMP stimulates neurite outgrowth in PC12 cells by a mechanism differing from those of NGF and bFGF.
