Following the intravenous injection (to the femoral vein) of <SUP>125</SUP>I-porcine proinsulin (6.2 μg (3.0 μCi) /Kg B.W.) in Zucker rats, the elution profile on G-50 Sephadex (fine grade) 0.9×22 cm column was obtained and compared with that obtained from normal Wistar rats, after periodical sampling of plasma in the circulation.<BR>In the Zucker rats, the Gel filtration patterns revealed a gradual increase in the radioactivity at the insulin position with the concurrent decrease in the proinsulin position after 15 minutes following the <SUP>125</SUP>I-proinsulin injection.<BR>Virtually no radioactivity was found at the insulin position up to 2 hours, in the case of normal rats. The radioactive material at the insulin position obtained from the Zucker rat was examined by the preparative polyacrylamid edisk gel-electrophoresis and electrophoretic behavior similar to monocomponent porcine insulin was confirmed.<BR>Blood sugar levels of both species of rat were lowest within 15 minutes and returned to the basal level within 60 to 120 minutes after the <SUP>125</SUP>I-proinsulin injection.<BR>These results suggest that the extrapancreatic conversion and degradation of proinsulin to insulin are present in the Zucker rats, which are assumed to require an additional insulin supply because of their insulin resistance. It should be mentioned that the different aspect (s) between the two species may be due to the different insulin clearance rate from the plasma under the same supply of proinsulin.
