Three human normal female groups aged 20-30, about 50 and over 60, and three pregnant groups of the first, second and the last trimester were included in the present studies. Each woman in these groups was injected intramuscularly with a single dose of ACTH-Z (20 units), and in the following two days, 17-OHCS content in the 24-hour urine was estimated by the Porter-Silber method.<BR>The results of experiments are summarized as follows : <BR>1) After injection of ACTH-Z, 17-OHCS, values of 24-hour urine reached in general at the peak on the first day, and a consequent drop to the preinjected values was observed on the second day.<BR>2) Responses to ACTH-Z injections were different among three groups of normal females. Average level of urinary 17-OHCS of normal females of 20±30 years of age was 3.505±0.250 mg/24-hrs. before injection of ACTH-Z, while the average value of 17-OHCS of the 1st 24-hoururine of the females of 20±30, about 50 and over 60 years of age were 14.50±0.803 mg, 9.501±0.297 mg and 8.008±0.589 mg, respectively.<BR>3) In pregnant females the urinary 17-OHCS excretions showed higher levels in the first trimester than nonpregnant females. In the second trimester the values were decreased slightly and in the last trimester the values were increased again.Following injection of ACTH-Z, the average urinary 17-OHCS value of the pregnant woment in the first trimester was 9.44±0.772 mg, the second trimester 5.482±0.734 mg and the last trimester, 8.888±1.396 mg per 24 hours, respectively. These data indicate a lower response of pregnant women to ACTH than that of nonpregnant.
