Many theories have been proposed regarding factors concerned with the mechanism of the regulation of aldosterone secretion. While the humoral agents e.g. ACTH, Adrenoglomerulotropin from pineal body, or pressure substance from kidney are suggested as stimulating factors, several authors denied the role of the nervous system in the stimulation of aldosterone secretion. But studies in our department during the past few years showed that there is a splanchnico-adrenocortical system beside the pituitary-adrenocortical axis. The effect of electrical stimulation of splanchnic nerve on aldosterone secretion was therefore investigated.<BR>Adult Mongrel dogs of approximately 10kg body weight were nsed. Operations were performed after subcutaneous administration of 2% solution of morphine hydrochloride at doses of 0.4ml per kg body weight as premedication, under local anesthesia with 2% procaine hydrochloride solution. Intravenous administration of preserved dog blood was used to maintain the blood pressure. Adrenal venous blood was obtained during one-hour periods, before and during stimulation through a canula inserted into the right lumbo-adrenal vein with translumber and retroperitoneal appreach. For the purpose of splanchnic nerve stimulation, S.W.D. 6-10 volt, 50 c/s, 5 m.sec. was applied for 10 minutes in bursts of 20 seconds on and 40 seconds off, followed by 10 minutes-intervals. This cycle was repeated three times.<BR>Silber-Porter's method was used for the determination of blood 17-OHCS, and a modified Neher & Wettstein's technic was used to estimate the adrenal venous blood aldosterone.<BR>Adrenal venous blood aldosterone was increased by splanchnic nerve stimulation in 4 of 6 cases, whereas no increase was observed in 4 control experiments. No changes were observed in adrenal blood output and 17-OHCS before and during stimulation in all animals.<BR>It is concluded that the splanchnic nerve plays a role in stimulating the synthesis or the secretion of aldosterone.
