Okamoto et al. have reported that the development of the Langerhans' islets is stunted in the offsprings descended from rabbits made diabetic on both paternal and maternal sides for the successive generations, and that the degree of the stunting is in accordance with the successive generations and spontaneous development of diabetes mellitus is observed in the rabbits of the F<SUB>4</SUB> and F<SUB>5</SUB>.<BR>The author carried out the following experiments. Namely, the F<SUB>4</SUB>rabbits of both sexes, which were descended from rabbits made diabetic with 5- (p-hydroxyphenylazo) -8-hydroxyquinoline on both paternal and maternal sides for the successive generations, developed “the spontaneous diabetes, ” and were mated over a month after their recovery from the diabetes mellitus, and thus rabbits “A” were begotten. Then “A” rabbits were mated and rabbits “B” were begotten, and these matings were repeated similarly to obtain rabbits “C” and “D”. The progress and the state of alloxan diabetes of these offsprings were also investigated. Some of the rabbits “A”, “B”, “C” and “D” sacrificed when they weighed over 2000g, 120 days after their birth, and histometrical studies of several organs were carried out. Comparing the measuring values obtained in the organs of normal, F<SUB>1</SUB>, F<SUB>2</SUB>, F<SUB>3</SUB>, and F<SUB>4</SUB> rabbits, the following results were obtained.<BR>1) There was no signiffcant difference in the rate of the weight increase among normal, A. B, C and D rabbits.<BR>2) Rabbits A, B, C and D were found to be always non-diabetic.<BR>3) On the histometry of the Langerhans' islets of the pancreas, the following results were obtained. (i) Stunted development of the islets was observed on the rabbit A, but the degree of the stunted development was less than of parents-the F<SUB>4</SUB> rabbits, being similar to that of the F<SUB>3</SUB> rabbit. (ii) Stunted development of the islets was also observed on the rabbit B, but its degree was less than that of parents-A rabbits and was similar to that of the F<SUB>2</SUB> rabbit. (iii) Stunted development of the islets was also observed on the rabbit C, but its degree was less than that of parents-B rabbits and was similar to the F<SUB>1</SUB> rabbit. (iv) No stunted development of the islets was observed on the rabbit D and the same value as that of normal rabbit was observed. (v) The number of B-cells of the islets on A, B, C and D increased approximately by five in accordance with each generation.<BR>4) As for the percentage of the cells in the anterior lobe of the pituitary gland, stepwise of eosinophilic cells and increase of neutrophilic cells and basophilic cells, were observed, and the values of A, B, C and D coincided those of the F<SUB>3</SUB>, F<SUB>2</SUB>, F<SUB>1</SUB> and normal rabbits respectively.<BR>5) Significant difference in the area of the follicle in the thyroid gland, the follicular lumina, the glandular cells of the submaxiliary gland, the liver cell and of the renal glomerulus could be found between the F<SUB>4</SUB> rabbit and the normal rabbit. On A, B, C and D rabbits, however, approximately the same value was observed as that observed on the normal rabbit.<BR>6) Long continuous diabetes mellitus was developed on A, B, C and D rabbits by the injection of alloxan in same dosage that could cause diabetes mellitus on the F<SUB>3</SUB>, F<SUB>2</SUB>, F<SUB>1</SUB> and normal rabbits respectively.<BR>7) Summarizing (3) - (6), the following conclusions could be made. When the offsprings from diabetic rabbits in 4 successive generations (F<SUB>4</SUB>) are mated under non-diabetic state in successive generations, the stunted development of Langerhans' islets is observed. Its degree, however, becomes always less than that of the parents.
