The role of β-glucuronidase in tissues is still not defined exactly and is a subject of controversy.<BR>The behaviours of mouse and rat tissue and rabbit serum β-glucurouidase activity in various experimental conditions such as hormone administration, castration, adrenalectomy, glucose administration etc. were observed.<BR>β-glucuronidase activity was determined by the modified method of Fishman Springer and Brunetti (1948).<BR>Results are summarized as followings ; <BR>1) In female mice, castration resulted in a marked decrease of uterine β-glucuronidase and the enzyme is returned to normal or higher level by estradiol administration.<BR>Progesterone showed no effect on the castrated uterine β-glucururonidase.<BR>2) In male mice, castration caused a reduction of the kidney β-glucuronidase activity. Testosterone propionate produced marked elevations in kidney β-glucuronidase activity in intact and castrated mice.<BR>In rat, testosterone propionate gave no effect upon the kidney β-glucuronidase.<BR>3) In rabbit, epinephrine and cortisone caused elevation in serum β-glucuronidase activity, norepinephrine and estradiol benzoate were less effective.<BR>Testostosterone propionate and thyradine injection caused no definit change in serum β-glucuronidase activity.<BR>Progesterone administration resulted in a decrease in serum β-glucuronidase activity.<BR>4) In rabbit, castration in either sex caused no expected change in serum β-glucuronidase activity and bilateral adrenalectomy produced marked reduction in it.<BR>5) Chlorpromazine and acetylcholine administration caused marked elevations in rabbit serum β-glucuronidase activity and the former produced increase in mouse kidney and muscle β-glucuronidase.<BR>6) By glucose, alloxan, and insulin administration, rabbit serum β-glucuronidase activity fluctuated in parallel with the change of the blood suger level. The fluctuation was more sensitive and uniform than that of glucuronidogenic hormone administration.<BR>7) From these results, though direct evidence to connect β-glucuronidase and glucose is lacking, glucose and related sugar metabolism may be one of the major components influencing the change of the β-glucuronidase activity, and glucuronidogenesis per se' is presumably not a major factor affecting the β-glucuronidase activity and elevation in the β-glucuronidase activity induced by hormone administration might be due to their action upon sugar metabolism.<BR>It is undoubtedly premature to make any conclusion about β-glucuronidase action in vivo and at present, it would be proper to consider the main action of β-glucuronidase is a hydrolytic one, and in some cases it may be associated with the change of the sugar metabolism.
