It has been demonstrated by Brecher & Waxler (1949) that the administration of goldthioglucose in mice would develop obesity. Later, Marshal (1955) found hypothalamic lesions in these mice. <BR>The present report deals with a marked effect of goldthioglucose (G.T.) on blood sugar in rabbits. <BR>Four rabbits, weighing 2.5 to 3.5 kg of body weight, received 100 mg of G.T. intramuscularly developed a marked hyperglycemia. The hyperglycemia occurred on the following day of injection and lasted for 1 to 2 weeks. However, the second injection of the same dose of C.T. given intramuscularly 1 to 2 months after the first injection failed to produce hyperglycemia. One of the four rabbits developed diabetes mellitus approximately 3 weeks after the second injection. In the rabbit, lesions of ventromedian nuclei and suprachiasmatic region of the hypothalamus, and atrophic changes of the pituitary and adrenal glands were demonstrated. Since no significant histological changes were observed in the pancreas, the cause of diabetes mellitus may be ascribable to the hypothalamic lesion.
