In order to clarify the effects of hemodialysis membrane on cytokine production, intra- and inter-dialytic changes of cytokine production from cultured mononuclear cells (MNC) were compared by crossover maneuver between ordinary cellulose membrane(OC) and polyethylene glycol chain grafted cellulose membrane (PC). 6 hemodialysis (HD) patients who underwent HD using OC for more than 2 weeks were examined first. And then membrane was changed to PC and more than 2 weeks later blood was drawn. MNC were separated and cultured with or without lipopolysaccharide(LPS) for 18 hours. Supernatant and cell lysate were collected and the concentration of TNFα, IL-1β, IL-6 in them were measured by ELISA. The total of supernatant and cell lysate was regarded as the production from INC. No significant differences were observed in pre-HD cytokine production between the two membrane periods except from less production of TNF α under unstimulated condition from MNC on OC period. On OC period, all cytokine productions were decreased at 30min after the start of HD, and IL-1β and IL-6 productions remained to be supressed until the end of HD. But on PC period, no significant changes were observed intradialytically. These results suggest that PC exerts less effects on MNC during HD than OC, however those difference affects little at least during short term observation.
