Using a freeze-substitution method, we examined light microscopic localizations of hydrolase activities in the alimentary canal of six species of parasitic copepods: Conchyliurus quintus, Modi-olicola bifidus, Ostrincola koe, Panaietis yamagutii, Neoergasilus japonicus and Ergasilus orientalis. Alkaline phosphatase activity was detected in the striated border of the non-vacuolar midgut cells. Acid phosphatase activity was detected in the secretary vacuoles of the vacuolar midgut cells and the striated border of the non-vacuolar midgut cells. Non-specific esterase, β-N-acetylglucosami-nidase and β-glucuronidase activities were detected only in the secretory vacuoles of the vacuolar midgut cells. No positive reactions were observed in the labral glands. These results suggest that digestive enzymes of these parasitic copepods are lysosomal enzymes secreted by the vacuolar midgut cells.
