We examined the susceptibility of Yamame embryonated liver cells, YEL-13, to Oncorhynchus masou virus, OMV. OMV did not produce syncytium on a YEL-13 cell sheet. Degeneration of YEL-13 was observed microscopically at the 9-12th day of infection and plaques were formed at the 18th day when incubated at 15°C, whereas syncytia on YNK were observed within 5 days and plaques were produced at the 7th day. The cytopathic effects (CPE) of OMV to YEL-13 were characterized by picnotic nucleus, basophilic cytoplasm, and round cell shape. At 20°C, no degeneration nor syncytium was observed both on YEL-13 and YEK cell sheet. When transferred from 20°C to 15°C, a few plaques with similar CPE to the cells incubated at 15°C from the beginning appeared in OMV-infected YEL-13 within a week. YEL-13 had about 10 times lower susceptibility to OMV and needed 2 or 3 times more time for visual plaque formation by OMV than YNK. As plaque assay of OMV in YEL-13 revealed one-hit kinetic pattern, a plaque on YEL-13 cell sheet must be initiated and produced by one particle of OMV.
