In the previous study, we reported about the measurement methods in the vertical diffusion method. In this report, the relationship between the bacilli species and the length of inhibition zones of the instilled drugs was studied.<BR>1) Five different quantities of B. C. G. were inoculated on 3% Ogawa media. After 2 days incubation in horizontal state, the 5 different doses of INH were poured into the bottom of the media and kept in vertical state for 4 weeks. It was found that the length of inhibition zone was the longer when the quantity of inoculum was smaller (Tab. 1 and Fig. 1).<BR>2) Directly after the inoculation of 0.05mg of 5 different species of mycobacteria (B. C. G., M. Fortuitum. H<SUB>7</SUB>, M. Smegmatis and M. Phlei) on 1% Ogawa media, Ethambutol and INH were poured each into the bottom of them in different doses. The incubating period was 3 weeks for B. C. G., 2 weeks for M. Fortuitum, and 4 days for the others. To measure the Ethambutol, M. Phlei was most suitable as seen in Fig. 2. Because by using this bacillus, not only the measurement was possible at a very low concentration of the drugs, but also the inclination of the inhibition zone by different doses was the slowest.<BR>For INH, B. C. G. was most suitable in the above meaning (Fig. 3). Thus, each drug has its most suitable bacillus for the vertical diffusion method.<BR>Next, we compared B. C. G. and H<SUB>37</SUB>Rv, but there was no difference between them (Fig. 4).<BR>3) The preservable period of B. C. G. was much influenced by the presence of NaOH. B. C. G. diluted by water was preservable at least for 10 days in the refrigerator or even at room temperature (winter), although the growth of bacilli became worse by 2 hours contact with 1% NaOH at room temperature (winter) and the length of inhibition zone became longer accordingly. When they were preserved for more than 2 days with NaOH at room temperature, growth of bacilli became so scanty that the inhibition zone could not be formed (Tab. 2).
