OKA-KATAKM1A medium, Ogawa 1% mono-potassium phosphate medium and Lowenstein-Jensen medium were compared with the potato flour added acid egg medium prepared by addition of potato flour 5.0g, whole egg 200.0 ml, glycerin 6.0 ml and 2% malachite green 6.0 ml into the original solution which was the mixture of monopotassiurn phosphate 1.0g, asparagine 0.5g and distilled water 100.0 ml. Quantitative cultivation of human type tubercle bacilli and dried BOG vaccine on the potato flour added acid egg medium showed much quicker development of greater number of colonies than the other three media. Dif terence in the number of colonies formed in each test tube inoculated with the same quantity of the inoculum was much smaller and the size of colonies formed was much greater than those of the other three media.<BR>Quantitative cultivation of sputum was made by Ogawa's method on Ogawa 3% mono-potassium phosphate medium and on potato flour added acid egg medium in which mono-potassium phosphate was increased to 3% of the original solution and asparagine was removed. Out of 67 total sputa cultured, 37 were positive in which 24 (64.9%) were positive on both media and 13 (35.1%) were positive only on potato flour added acid egg medium, and there was not a single case of positive only on Ogawa medium. Moreover, the medium added with potato flour showed earlier development of more colonies with larger size similar to the findings of the quantitative cultivation of the bacterial suspension.
