The effects of sodium fluorde on the growth and protein synthesis of HeLa cells and fluoride-resistant cells was studied. Fluoride-resistant cells of HeLa cells were developed by progressively increasing the concentration of sodium fluoride in the growth medium to 2 mM sodium fluoride, the concentration of sodium fluoride at which the growth of HeLa cells was arrested completely. Protein synthesis (incorporation of <SUP>14</SUP>C-leucine) was markedly reduced for a 24-hr period after the addition of 2 mM sodium fluoride in HeLa cells, but not affected in fluoride-resistant cells.<BR>The changes in newly synthesized cytoplasmic proteins of HeLa cells and fluoride-resistant cells after sodium fluoride treatment were investigated by sodium dodecyl sulfate polyacrylamide gel electrophoresis followed by autoradiography. The peptide pattern of both HeLa cells and fluoride-resistant cells obtained from stained gel were identical. In the presence of sodium fluoride, HeLa cells synthesized one particular peptide, the molecular weights of this peptide being about 77000 daltons. The synthesis of this 77KD peptide increased with the duration of time of sodium fluoride treatment, and this peptide was detected in plasma membrane, microsomal fraction and mitocomdrial fraction, but not in nuclei or supernatant fraction.
