A new convenient method for assaying dextransucrase is described. The method is based on the enzymatic conversion of soluble Blue Dextran 2000 to a water-insoluble form in the presence of sucrose. The water-insoluble product was collected on a glass fiber filter and resolubilized in a NaOH solution to determine the absorbance at 620 nm, at which Blue Dextran 2000 shows maximum absorbance. The absorbance at 620 nm increased linearly with the enzyme concentration and the reaction time an initial lag. The amount of insolubilized Blue Dextran showed a proportional relationship with that of the glucosyl moiety incorporated into the insoluble product from sucrose measured by radioactivity. The method was applicable to various preparations from the crude extract to the purified enzyme. The role of Blue Dextran in the enzyme reaction is also discussed.
