Lipomyces starkeyi ordinarily forms an intracellular oil globule and oil globule markedly develops during the retardation phase of growth (from 40 th to 90 th hr of culture period). The effect of iron concentration (0.04mg/l, 0.1mg/l, 0.5mg/l, 1.0mg/l) in chemically defined media both on the growth pattern and on the development of the intracellular oil globule in the yeast was examined. Development of the oil globule was able to be expressed as the amount (mg) of total lipids extracted from 108 cells (total lipids/108 cells), because the variation profile of the volume of the oil globule as a function of culture time agreed well with that of the total lipids/l08 cells at various concentrations of iron. When iron was at a concentration of 0.04mg/l, the apparent growth (O. D. 660) and the total lipids/108 cells were the lowest over the retardation phase of growth. When the iron concentration increased to 0.1mg/l and 0.5mg/l, the apparent growth and the total lipids/108 cells increased. But when the concentration of iron was 1.0mg/l, the total lipids/108 cells decreased. On the other hand, total cell number in the media was not affected at any phase of growth by the variation of initial concentration of iron. From these results, we concluded that the increase of the iron concentration in the medium first causes the increase of the total lipids/108 cells and second, as a direct result of this, leads to the increase of the apparent growth of cells. Under the conditions tested, most part of iron added in media transferred to cell fractions during the logarithmic phase of growth and the transfer completed at a middle retardation phase of growth. These results suggest that cellular activities for the total lipids synthesis in L. starkeyi were affected by iron concentration of cell fractions.
