In the hydrolysis of saponin in the tubers of Dioscorea tokoro M., enzymatic hydrolysis with several Aspergillus and Penicillium species were compared with known acid hydrolysis. Aspergillus terreus D4 was selected as the most favorable strain, and with this strain, a novel, effective, and semi-industrial solid culture method was developed. Hydrolysis was repeated twice using the combination of enzyme-enzyme or enzyme-acid. First hydrolysis with enzyme gave about 90_??_95% of diosgenin, and second hydrolysis with enzyme increased the yield of diosgenin to about 108%, while that with acid to about 120% as compared with the known method of acid hydrolysis.
