The effects of prostaglandin E1 (PGE1) on mucin secretion and mucosal lesions in rat stomach induced by indomethacin and cold restraint were investigated.The mucus content was measured by the alcian blue binding method. Intravenous infusion of PGE1 (10μg/kg) for 1 hour increased the total amount of mucosal and adhesive mucus at the rate of 14.2%. The rate of increase in the amount of adhesive mucus on the surface epithelium was 16.7%. Both treatment by oral administration of indomethacin (60mg/kg) and by cold restraint at 4 °C for 1 hour produced acute mucosal lesions and decreased the mucus contents in gastric mucosa at the rate of 17.8% and 16.1%, respectively. However, the mucosal lesions and decreases in mucus content were markedly inhibited by the administration of PGE1 (10μg/kg) for 1 hour infused simultaneously with the above treatments.These results suggest that a concentration of PGE1 so low that it hardly inhibited gastric acid secretion could induce an increase in mucin secretion and prevent ulcer formation in rat stomach.
