:I. Microassay of Cyclic Nucleotides in Rat Stomach

We designed a microassay for cyclic nucleotides using a modification of Lowry's quantitative histochemical approach. Levels of cyclic AMP and GNP were simultaneously assessed in 500μg dry weight tissue from the stomach area. These tissues included gastric mucosa, propria muscles in the forestormach and the fundic and pyloric regions. After deproteinization, lyophilization and succinylation, 40μl of tissue extractions were radioimmunoassayed to determine the levels of cyclic AMP and GMP. The recovery rates were 88.9±1.2% and 88.6±0.8, respectively. In the mucosa, the contents of these nucleotides in the pyloric region were significantly high as compared with the other two regions (p<0.001). The level of cyclic AMP was highest in the forestomach and the values were statistically significant compared to findings in the fundic region (p<0.01). Thus, these two nucleotides apparently have different functions in the different gastric regions.
